چکیده

Abstract Introduction Central nervous system (CNS) metastases are devastating neurologic complications. However, the clinical value of cerebrospinal fluid (CSF) as liquid biopsy medium in EGFR-mutated or ALK-rearranged patients with brain (BM) leptomeningeal (LM) remains unclear. Patients and Methods We retrospectively analyzed gene map 14/48 NSCLC-BM 34/48 NSCLC-LM patients. Compare predictive CSF matched plasma (PLA) for single CNS progression (cohort 1) intracranial simultaneously extracranial disease 2). Circulating cell-free tumor DNA (ctDNA) PLA underwent next-generation sequencing targeting 168 cancer-relevant genes. Results For progression, mutation rate major driver genes is much higher than PLA. Besides, also demonstrated levels potential prognostic markers comparison samples, showing EGFR-amp presented more two times compared to PLA, detection TBX3, PTEN were almost double paired samples. In addition, poor outcomes associated makers including CDKN2A/B, PIK3CA/G, CDK4/6, MET showed exclusively samples.Almost all mutations identified could detect samples both progression. rates mutations, consisting EGFR ALK these survival marker like EGFR-amp, MET, PIK3CA uniquely Conclusion comprehensive genomic profile NSCLC -LM regardless evolution.

Cerebrospinal fluid-derived circulating tumour DNA better represents the genomic alterations of brain tumours than plasma

Cell-free circulating tumour DNA (ctDNA) in plasma has been shown to be informative of the genomic alterations present in tumours and has been used to monitor tumour progression and response to treatments. However, patients with brain tumours do not present with or present with low amounts of ctDNA in plasma precluding the genomic characterization of brain cancer through plasma ctDNA. Here we s...

Detection of tumor-derived DNA in cerebrospinal fluid.

The sensitivity of PCR-based methods for the detection of DNA offers opportunities for tumor diagnosis from the small amounts of tumor-derived DNA released into body fluids. We report the detection of tumor DNA in the cerebrospinal fluid (CSF) of two patients with intracranial neoplasms. One patient had a metastatic breast carcinoma which contained amplified HER-2/neu genes, and amplified HER-2...

Circulating tumor-derived DNA is shorter than somatic DNA in plasma.

Circulating tumor DNA (ctDNA) is now widely investigated as a biomarker in translational and clinical research (1). However, despite the growing field of clinical applications, the biology of ctDNA remains unclear. In trying to learn about the origins of ctDNA, nature provides us with very few clues. One of the important accessible parameters is the size of those DNA fragments. In addition, a w...

Diagnostic value of circulating tumor cells in cerebrospinal fluid

OBJECTIVE To assess circulating tumor cells in cerebrospinal fluid as a diagnostic approach to identify meningeal metastasis in patients with non-small cell lung cancer by using tumor marker immunostaining-fluorescence in situ hybridization (TM-iFISH). METHODS In 5 non-small cell lung cancer patients who were confirmed to have developed meningeal metastasis by cerebrospinal fluid cytology, 20...
